Long-term experimental sites are expected to provide important information regarding soil properties as affected by management practices. This study was designed to examine the effects of continuous fertilization, and manuring on the activities of enzymes involved in mineralization of C, N, and P on a long term (33 years) field trial under sub-temperate conditions in India. Treatments at the site included application of recommended doses of nitrogen and phosphorus (NP), nitrogen and potassium (NK), nitrogen, phosphorus and potassium (NPK), farmyard manure (FYM) with N (N + FYM), FYM with NPK (NPK + FYM) and un-amended control (C). The study was done under rainfed soybean–wheat rotation. Manure application increased soil carbohydrate, dehydrogenase, acid and alkaline phosphatases, cellulase, and protease activity significantly. Urease activity was not influenced by the manure treatment and the activity was highest in controls. Both acid and alkaline phosphatase activities were negatively influenced by chemical fertilizer treatment. Almost all the enzymes studied were significantly correlated with soil C content. The results suggest that application of FYM directly or indirectly influences the enzyme activity and it in turn regulates nutrient transformation.  相似文献

Previous studies describe the suitability of a new type of phosphorus (P) fertilizer, called “rhizosphere‐controlled fertilizer” (RCF), to supply available P to plants while reducing soil phosphorus fixation. In order to explore the involvement of organic acid root exudation in P uptake from RCF, we investigated the relationship between shoot and root P concentrations, and the concentration of the main polycarboxylic organic acids in roots, shoots, and plant exudates. Plant species with different P‐acquisition efficiency (low: maize; medium: chickpea; high: lupin) were grown in hydroponics with three different P fertilizers: The water‐insoluble P fraction of RCF (RCF); Phospal, a slow‐release source of phosphate composed of calcium and aluminum phosphates (PH); monopotassiumphosphate (KP), and a control treatment without P (P–). RCF was as efficient as KP in supplying P to plants in the case of chickpea and lupin, and slightly less efficient than KP in maize. However, P from PH was not available for maize and less available compared to KP and RCF in chickpea and lupin. This variation reflects the different efficiencies in P acquisition for the three plant species. Except in the case of maize, plants receiving KP presented the lowest concentration of organic acids in roots and exudates, while those plants suffering severe P deficiency (P– and PH) showed the highest organic acid concentration. However, RCF had a high concentration of organic acids in roots and exudates, as well as a high P concentration in the shoot indicating that P uptake from RCF is enhanced due to root release and action of specific organic acids.  相似文献

Untreated oil palm empty fruit bunch (REFB), alkali treated EFB (AEFB), ultrasound treated EFB (UEFB) and simultaneous ultrasound-alkali treated EFB (UAEFB) short fibers were incorporated in poly(lactic acid) (PLA) for fabricating bio-composites. The REFB fiber-PLA (REPC) and treated EFB (TEFB) fiber-PLA (TEPC) composites were prepared and characterized. Glass transition temperature, crystal melting temperature, decomposition temperature, melt flow index, density and mechanical properties (tensile strength, tensile modulus and impact strength) of TEPC are found to be higher than those of REPC. The observed crystallization temperature of TEPC is lower than that of REPC. Among all samples, TEPC prepared from UAEFB fiber shows better performances than other samples fabricated by REFB and AEFB fibers. Scanning electron microscopy, Fourier transform infrared spectroscopy and XRD analyses well support all the observed results.  相似文献

Background:Some resources have suggested that genetically inactivated PTs bear a more protective effect than chemically inactivated products. This study aimed to produce new version of PT, by cloning an inactive PTS1 in a fusion form with N-terminal half of the LLO pore-forming toxin. Methods:Deposited pdb structure file of the PT was used to model an extra disulfide bond. Codon-optimized ORF of the PTS1 was used to make recombinant constructs of PTS1 and LLO-PTS1 in the pPSG-IBA35 vector. The recombinant PTS1 and LLO-PTS1 proteins were expressed in BL21 DE3 and SHuffle T7 strains of E. coli and purified by affinity chromatography. Cytotoxic effects of the recombinant proteins were examined in the MCF-7 cell line. Results:The purity of the products proved to be more than 85%, and the efficiency of the disulfide bond formation in SHuffle T7 strain was higher than BL21 DE3 strain. No cytotoxicity of the recombinant proteins was observed in MCF-7 cells. Soluble recombinant PTS1 and LLO-PTS1 proteins were produced in SHuffle T7 strain of E. coli with high efficiency of disulfide bonds formation. Conclusion:The LLO-PTS1 with corrected disulfide bonds was successfully expressed in E. coli SHuffle T7 strain. Due to the safety for human cells, this chimeric molecule can be an option to prevent pertussis disease if its immunostimulatory effects would be confirmed in the future. Background:

Ovarian tissue cryopreservation is an alternative strategy to preserve the fertility of women predicted to undergo premature ovarian failure. This study was designed to evaluate the expression of folliculogenesis-related genes, including factor in the germline alpha (FIGLA), growth differentiation factor-9 (GDF-9), follicle-stimulating hormone receptor (FSHR), and KIT LIGAND after vitrification/warming of human ovarian tissue.

Methods:

Human ovarian tissue samples were collected from five transsexual women. In the laboratory, the ovarian medullary part was removed by a surgical blade, and the cortical tissue was cut into small pieces. Some pieces were vitrified and warmed and the others were considered as non-vitrified group (control). Follicular normality was assessed with morphological observation by a light microscope, and the expression of FIGLA, KIT LIGAND, GDF-9,, and FSHR genes was examined using real-time RT-PCR in both the vitrified and non-vitrified groups.

Results:

Overall, 85% of the follicles preserved their normal morphologic feature after warming. The percentage of normal follicles and the expression of FIGLA, KIT LIGAND, GDF-9, and FSHR genes were similar in both vitrified and non-vitrified groups (P > 0.05).

Conclusion:

Vitrification/warming of human ovarian tissue had no remarkable effect on the expression of folliculogenesis-related genes. Key Words: Vitrification, Gene expression, Humans  相似文献

To explore long-term impact of organic and inorganic fertilizers on soil health and grain quality, we monitored the enzyme activities and chemical properties of soil; and chemical composition of grain from eight treatments at an experimental field site established in 1996. There were eight treatments applied to both wheat and maize seasons: a control; four inorganic fertilizers, that is, nitrogen and phosphorus (NP), nitrogen and potassium (NK), phosphorous and potassium (PK) and nitrogen, phosphorus and potassium (NPK); farm yard manure alone (FYM) and addition of FYM at two different doses (100 and 50% of recommendation) to NPK that is, NPK + FYM and ½ NPK + FYM. After 11 years of the experiment the NPK + FYM and ½ NPK + FYM treatments had the highest yields, about 5 Mg maize ha−1 and 2 Mg wheat ha−1 with about 2 and 0.5 Mg ha−1, respectively more than the NPK treatments. The dehydrogeanse activity of soils increased significantly in FYM and ½ NPK + FYM. Except urease all other enzymatic activities were increased in those treatments, which received manure. Urease activity was higher in mineral-N applied plots. Grain protein content of both maize and wheat was highest in mineral fertilized plots. Test weight also increased significantly on application of mineral fertilizer. Plots treated with half dose of recommended mineral fertilizer along with FYM were higher in urease, phosphomono and diesterase activities than that of NPK + FYM treated plots. Long-term application of inorganic nutrients along with FYM improved grain mineral composition and yield. Inhibition of few enzymatic activities were also observed upon application of inorganic nutrients either alone or in combination.  相似文献

OBJECTIVE: To compare intake estimates, validity and reliability of two summary questions to measure fish consumption with information from a detailed semi-quantitative food-frequency questionnaire (FFQ) on fish consumption. DESIGN: Population-based, cross-sectional study. Participants completed an FFQ and provided blood samples for erythrocyte membrane eicosapentaenoic acid (EPA) analysis. Aggregate measures of consumption of fresh/frozen/canned fish (fresh fish) and smoked/salted/dried fish (preserved fish) were generated from the FFQ and were compared with responses to the summary questions regarding intakes of similar items. Both methods were tested for validity, using correlation and linear regression techniques with EPA, and retest reliability. SETTING: Perth metropolitan area, Western Australia. SUBJECTS: One hundred and nine healthy volunteers of both sexes, aged 21-75 years. RESULTS: The summary fresh fish measure underestimated frequency and grams per week given by the aggregate question by about 50%, while estimates from the summary preserved fish measure were approximately three times that of the aggregate measure. Multiple linear regression analysis suggested that the aggregates accounted for more of the variation in EPA levels, but the difference was minimal. Intra-class correlations confirmed that both methods were reliable. CONCLUSIONS: Our study indicates that extensive questioning results in different absolute intakes of fish compared with brief questioning, but does not add any information if ranking individuals according to overall consumption of fish.  相似文献
